Applications

AA-Ac was developed for rapid profiling of complex oligosaccharide mixtures [Charlwood et al (2000)].

AA-Ac labeled oligosaccharides can be analysed by HPLC, MALDI-MS, free-zone capillary electrophoresis, and capillary electrophoresis / electrospray ionization mass spectrometry [Charlwood et al (2000)].

Notes

AA-Ac is a versatile fluorescent probe for glycan analysis. It is superior to its predecessor AMAC (2-aminoacridone) with regard to fluorescence intensity and replaces AMAC for many applications. The latter probe has been used in a variety of studies including those on human milk glycans [Charlwood et al (1999(], human transferrrin [Charlwood et al (1998)], immunoglobulins [Okafo et al (1996)], and glycosylaminoglycans [Kitagawa et al (1995)].

The LudgerSep N1 column (Cat # LS-N1-4.6x250) is an excellent choice for amide HPLC profiling and fingerprinting of complex AA-Ac-labeled glycan mixtures.

Kit Specifications

The kit contains two sets of the following reagents (supplied in glass ampoules sealed under pure nitrogen):

  • 3-(acetylamino)-6-aminoacridine (AA-Ac dye)
  • Dimethyl sulfoxide (DMSO)
  • Acetic acid
  • Sodium cyanoborohydride

Purity: > 99% by HPLC. Soluble in DMSO and ethanol. M.W. 278. lambda-ex = 382 or 445 nm, lambda-em = 525 nm.
